Duloxetine is a selective serotonin and norepinephrine reuptake inhibitor (SSNRI). The exact mechanism of duloxetine's antidepressant and central analgesic effects is not yet clear, but it is believed to be related to its enhancement of central nervous system serotoninergic and noradrenergic functions. The results of preclinical studies have shown that duloxetine is a strong inhibitor of neuronal serotonin norepinephrine reuptake, and its inhibitory effect on dopamine reuptake is relatively weak. In vitro research results show that duloxetine has no obvious affinity with dopaminergic receptors, adrenergic receptors, cholinergic receptors, histaminergic receptors, opioid receptors, glutamate receptors, and GABA receptors. Duloxetine does not inhibit monoamine oxidase.
