Tadalafil is a phosphodiesterase 5 inhibitor, originally developed by GlaxoSmithKline and later jointly developed by ICOS and Eli Lillv. It was approved by the FDA in 2003 as a treatment for male erectile dysfunction (MED). ) Drugs were marketed in the United States, and later it was discovered that another clinical application of tadalafil is the treatment of pulmonary hypertension. Tadalafil has a stronger effect on PDE5 than other phosphodiesterase enzymes. The effect of tadalafil on PDE5 is more than 10,000 times stronger than the effect on PDE1, PDE2, and PDE4 found in the heart, brain, blood vessels, liver and other organs. The effect of tadalafil on PDE5 is more than 10,000 times stronger than the effect on PDE3 found in the heart and blood vessels. Compared with similar drugs, tadalafil has the advantages of high selectivity, long half-life, and greater autonomy for patients.
