Originally researched as Pfizer, it was approved for marketing by the US FDA in 2005. Pregabalin is the first drug approved by the US FDA for diabetic peripheral neuropathy and post-herpetic neuralgia, and is currently the best-selling analgesic. Pregabalin is an analogue of γ-aminobutyric acid (GABA), similar in structure and function to gabapentin, and has antiepileptic, analgesic and anxiolytic activities. The antiepileptic mechanism of pregabalin is still unclear. In laboratory studies, pregabalin has anticonvulsant activity on various epilepsy models; the activity spectrum of animal models is similar to that of gabapentin, but the activity of pregabalin is 3-10 times that of gabapentin. In view of its superior clinical efficacy, in 2007 the drug was named one of the "Top Ten Medical Advances in 2007" by Time Magazine. Since its launch in 2004, its global sales have continued to rise. Its sales in 2018 were US$5 billion, making it one of the world's best-selling prescription drugs and a "blockbuster" drug.
