Doxylamine is the first-generation antihistamine, which can block histamine 1 (H1) receptors in a competitive, reversible, and non-specific manner. It can also block central or peripheral muscarinic receptors in a non-specific manner and has significant anticholinergic activity. As an alcoholic antihistamine, it can be used as a short-term sedative and hypnotic, or often used in compound prescriptions, and is often used as a short-term treatment for insomnia. As of 2004, doxylamine and diphenhydramine are the drugs most commonly used to treat short-term insomnia. Doxylamine combined with vitamin B6 can prevent morning sickness in pregnant women. The fetal safety grade of this drug is "A". The antiemetic effect of doxylamine is related to the antagonism of central cholinergic receptors and central H1 receptors.
