Clofarabine is a nucleotide analog. It was developed by HOFFMANN LA ROCHE and was approved by the US FDA in December 2004. Its trade name is Clolar. It is the first new drug approved for the treatment of childhood ALL in the past 10 years. Clofarabine is a second-generation purine nucleoside derivative. After it is phosphorylated into triphosphate by deoxycytidine kinase, it can effectively inhibit nucleotide reductase and terminate DNA synthesis, and it can also inhibit DNA polymerase. a, make the DNA chain no longer extend.
