Tetrabenazine is a safe and effective drug for the treatment of various hyperkinesis disorders. Compared with traditional antipsychotics, it does not cause tardive dyskinesia. Tetrabenazine is a compound known in the 1950s. Until 2008, the FDA approved it for the treatment of Huntington's disease with rapid approval qualifications, becoming the first and only drug for the treatment of Huntington's disease (HD) in the United States. Subsequently, Auspex developed the world’s first deuterated drug, deuterated tetrabenazine, based on the kinetic isotope effect (kH/kD=6.5). The incorporation of deuterium will reduce the rate of drug metabolism and the frequency of administration. The safety and effectiveness of the drug are significantly improved. In April 2017, the FDA approved the listing of deuterated tetrabenazine, becoming the first deuterated drug to be marketed in history. Deuterated tetrabenazine was included in China's first batch of clinically needed overseas new drugs in November 2018, and its marketing application was accepted by CDE in December 2019. If it is successfully approved for marketing, the world's first deuterated drug will also become China's first deuterated drug, which will definitely promote the development of deuterated drugs in China.
