Corbirestat is a new CYP3A inhibitor developed by Gilead and was approved by the US Food and Drug Administration on September 24, 2014. It is currently the only approved cocktail therapy AIDS drug enhancer. At present, in addition to unilateral, it is also used in combination with other anti-reverse transcriptase drugs for HIV-1 infection. In the Chinese Guidelines for the Treatment of AIDS, the recommended regimen for newly-treated adults and adolescents is 2 NRTIs backbone drugs combined with the third class of drugs. The third class of drugs can be NNRTIs or enhanced PIs (including ritonavir or cobipristat) ). Cobirestat itself has no anti-HIV activity, but it can increase the required concentration of anti-HIV drugs by inhibiting CYP3A, the main enzyme that metabolizes drugs in the human body. Corbirestat has been proven in clinical trials to be no less effective than ritonavir and has similar safety to ritonavir. Since its launch, darunavir tablets (anti-AIDS compound tablets composed of darunavir and cobirestat) have had sales of more than US$1 billion for five consecutive years. The US FDA has approved a number of anti-AIDS compound preparations containing cobirestat, and the compound anti-AIDS drugs containing cobirestat are expected to be popular in the future anti-AIDS drug market.
