Applications

Macitentan was developed by Actelion and was approved by the US FDA on October 18, 2013, for the treatment of pulmonary hypertension. Macitentan is an endothelin receptor antagonist. It can act on endothelin (ET) and its receptors that have changed functions in patients with pulmonary hypertension. It has dual effects on ET-A and ET-B receptors. Inhibition and tissue targeting can prevent excessive vasoconstriction, thereby alleviating disease symptoms and improving the quality of life of PAH patients. Therefore, macitentan has important clinical significance as an endothelin receptor antagonist. Compared with bosentan, macitentan has better tissue distribution, a higher affinity for non-selective endothelin (ET) receptors, fewer drug interactions, drug tolerance, and safety People are satisfied, so it has become a new type of medicine for PAH treatment. At present, it has been listed in more than 30 countries and regions around the world. Since its listing, the sales of macitentan have grown rapidly. In 2018, its sales almost doubled to US$2.573 billion.
